Justification

In its active form, Factor V is a cofactor, which together with activated Factor X, forms a prothrombinase complex that more efficiently converts prothrombin to thrombin than FXa alone. Congenital deficiency of Factor V leads to Owren’s disease (or parahemophilia), a rare inherited disorder that causes mild to severe bleeding. Deficiencies in Factor V activity may also be acquired secondary to other diseases such as liver disease, disseminated intravascular coagulation (DIC), or the development of a specific factor inhibitor.

DESCRIPTION This is a clot-based assay for the determination of Factor V Activity level in citrated plasma. Patient sample at three different dilutions is mixed with Factor V Deficient plasma containing normal levels of all other factors, such that the resulting Prothrombin Time is dependent only on the level of Factor V present in the patient sample. This value is compared to a reference curve generated from calibrators with known levels of Factor V Activity.
